Mosley proceeded to  play for the Alabama Crimson Tide in all 13 games in his first year in college and received Rivals.com, College Football News and Football Writers Association of America (FWAA) Freshman All-American honors. His second season was not so great as it was marred  by an elbow injury, he sustained in the fourth game of the season.

During the junior year, Mosley had 107 tackles, the highest at the time for any Alabama Crimson Tide player. He was  then shortlisted for the 2012 Butkus Award, where he emerged third, this earned him the  Alabama University football team MVP and he was honored with the first-team All-American by the Associated Press. In his final season of college football, the American born  once again was in top form, leading his team in tackles with 108 tackles. Baltimore Ravens (2014–2018)

At the time the  2014 NFL Draft was carried out, the Baltimore Ravens selected Mosley as its 17 overall-pick. He then  signed a four-year rookie contract worth $8.87 million with a signing bonus of $4.71 million.

Mosley was given the #57 jersey and named as one of the teams starting inside linebackers. thus making him the first Baltimore Ravens rookie to start for the franchise since 2006. After his first  debut for the Ravens, Mosley continued in top form, thus becoming  the first Ravens player to be selected to the Pro Bowl in their rookie season, when he was named to the 2015 Pro Bowl. He was again shortlisted for the NFL Defensive Rookie of the Year award, but he finished as a runner-up. He finished his first season in the NFL with 133 tackles. He then went on to the playoffs, where the Ravens defeated the Pittsburgh Steelers in the AFC Wildcard, and lost in the AFC Divisional Round to the New England Patriots.

He sustained a hamstring injury which put him out of play for a while, this included  two games and that was the first time he would not be part of a game start. In all 14 games played in the 2017 season, Mosley started them all and recorded 92 tackles. Following the completion of his rookie contract, Baltimore Ravens gave Mosley a base salary of $8.71 million for the 2017 season. Once again, the American born was named as a starting inside linebacker. He earned his third Pro Bowl selection and had 132 tackles in 16 games. Mosley was ranked 98th in the Top 100 NFL Players of 2018. In the 2018 season, Mosley once again earned another selection to the Pro Bowl. He had 105 tackles, the highest in the Baltimore Ravens team.

New York Jets (2019–date)

March 2019, saw Mosley sign a five-year deal with the New York Jets worth $85 million. He continued playing in top form with  the Jets but soon  sustained a groin injury in December. He thus became the 15th Jets player to be placed in the Injury Reserve list that season. It was reported that he will have to undergo surgery.
